Passive systemic anaphylaxis (PSA) induced by IgE is an established model for the study of anaphylaxis. This model is induced by the systemic injection of specific IgE antibodies into mice, followed by challenging with the corresponding antigen 24 h later. Here, we generated a PSA model using human immune system reconstituted HuHSC-NCG-M mice constructed by infusing human hematopoietic stem cells(HSCs) into the immunodeficient mouse NCG-M (strain No.T036669).

PCA model strategy is shown in Fig. 1A. Rectal temperature of mice were measured prior to injecting NP-KLH via the caudal vein. After the injection, the rectal temperature of the mice was measured every 10 minutes. Rectal temperature in the PSA model group started decreasing within 10 minutes post-challenge, while treatment of the PSA mice with omalizumab alleviated hypothermia (Fig.1B).
