UOX mouse phenotype and efficacy validation
1. The serum uric acid levels in Uox-KO mice were significantly increased compared with B6 mice. Allopurinol intervention can significantly decrease the serum uric acid levels in Uox-KO mice, with no impact on body weight.
2. At the end of 16 weeks of age, the kidney tissues of mice in each group were taken for pathological staining (H&E, Masson). The results show that Uox-KO mice showed significantly increased kidney injury compared with C57BL/6 mice, as shown by inflammatory cell infiltration (yellow arrow), extensive glomerular atrophy (red arrow), and cystic dilatation of the renal pelvis and calices (black arrow). What's more, Uox-KO mice showed hyperplasia of interstitial fibrous tissue and exudation of homogeneous red-stained material were observed in a small number of renal tubules compared with C57BL/6 mice.
